Abstract

Objective: To investigate the effect of Liuwei Dihuang Pill on the number, the surface marker, cell cycle and colony formation of HSC from mouse marrow.

Methods: Old Kunming mice were randomly divided into 4 groups: the control group, low, middle and high dose of Liuwei Dihuang Pill group. Then we separated the HSC from marrow after 7 days fed with saline or Liywei Dihuang Pill respectively, numerated monocyte, detected the surface marker and cell cycle of the HSC by FACS and tested the colony forming by semisolid media culture.

Results: Among the four groups, there was no obvious difference in the number of MNC, suspended cell and colony. The expression of Sca-1 and CD34 increased in the low and middle dosage group, it meant that the number of HSC elevated by low and middle dosage medicine. The ability of cell proliferation was also higher in the three dosage groups.

Conclusion: Liuwei Dihuang Pill activates HSC by increasing the number, proliferation and function of more primitive HSC.
