The aim of the study was to evaluate the functional properties of muffins fortified with white clover flowers (Trifolium repens L.), which were added to the dough in the following amounts: (i) 0% (control); (ii) 2.5%; (iii) 5.5%; (iv) 7.5%; and (v) 10%. The organoleptic properties were assessed by a panel of consumers. Additionally, the following parameters were also tested: basic chemical composition, total polyphenols, the antioxidant activity together with antiproliferative effects on the A375 melanoma cell line, starch nutritional fractions and the in vitro glycemic index. As a result, replacing wheat flour with white clover flour significantly affected the color, aroma and taste of the muffins. The content of proteins, fats, total ash, dietary fiber, resistant starch (RS), slowly digestible starch (SDS),total polyphenols and antioxidant activity increased statistically significantly with the elevated amount of white clover flour added to the dough. At the same time, the content of free glucose (FG), rapidly available glucose (RAG) and rapidly digestible starch (RDS), the value of the in vitro glycemic index and the viability of melanoma cancer cells decreased significantly. Swahili coastal forests, spanning the Kenyan and Tanzanian coastlines and the Zanzibar Archipelago, are integral to the biodiverse Eastern Arc and Coastal Forest region. These forest ecosystems face considerable anthropogenic threats. This study contributes to the available knowledge on the floral composition and plant community assemblages of Mnemba Island, a small sandy cay in the Zanzibar Archipelago, which helps to establish baseline data for conservation purposes. Two main coastal forest plant communities were identified: (1) the Casuarina cunninghamianaSuriana maritima open to closed sandy dry coastal forest, and (2) the Eugenia capensisMimusops obtusifolia coastal forest. A total of 91 different plant species belonging to 54 plant families were identified for the island. Community 2, an indigenous forest, supports diverse bird breeding colonies and is a crucial habitat for the threatened Aders’ duiker. Understory development is limited due to the historical Suni antelope overpopulation. Environmental factors like salt spray, allelochemicals, herbivore browsing, and climate fluctuations influence the vegetation abundance and composition. The study underscores differences in species diversity and composition between the planted Casuarina community and the natural atoll vegetation. The natural vegetation shows affinities with the Zanzibar–Inhambane edaphic coral-rag scrub forest and the transitional rainforest. Several species characteristic of Indian Ocean atolls were identified, providing insights into invasion ecology and conservation strategies. The study contributes conceptually to our understanding of vegetation dynamics in island ecosystems by highlighting the interplay between plant communities, environmental processes, and human activities. The fragile yet resilient nature of Mnemba’s unique ecosystem is emphasised, offering insights for conservation management, long-term monitoring, and adaptive approaches tailored to island environments. Integrating Large Language Models (LLMs) into e-government applications has the potential to improve public service delivery through advanced data processing and automation. This paper explores critical aspects of a modular and reproducible architecture based on Retrieval-Augmented Generation (RAG) for deploying LLM-based assistants within e-government systems. By examining current practices and challenges, we propose a framework ensuring that Artificial Intelligence (AI) systems are modular and reproducible, essential for maintaining scalability, transparency, and ethical standards. Our approach utilizing Haystack demonstrates a complete multi-agent Generative AI (GAI) virtual assistant that facilitates scalability and reproducibility by allowing individual components to be independently scaled. This research focuses on a comprehensive review of the existing literature and presents case study examples to demonstrate how such an architecture can enhance public service operations. This framework provides a valuable case study for researchers, policymakers, and practitioners interested in exploring the integration of advanced computational linguistics and LLMs into e-government services, although it could benefit from further empirical validation. Large language models (LLMs) can store factual knowledge within their parameters and have achieved superior results in question-answering tasks. However, challenges persist in providing provenance for their decisions and keeping their knowledge up to date. Some approaches aim to address these challenges by combining external knowledge with parametric memory. In contrast, our proposed QA-RAG solution relies solely on the data stored within an external knowledge base, specifically a dense vector index database. In this paper, we compare RAG configurations using two LLMs—Llama 2b and 13b—systematically examining their performance in three key RAG capabilities: noise robustness, knowledge gap detection, and external truth integration. The evaluation reveals that while our approach achieves an accuracy of 83.3%, showcasing its effectiveness across all baselines, the model still struggles significantly in terms of external truth integration. These findings suggest that considerable work is still required to fully leverage RAG in question-answering tasks. Full article

This study introduces a novel approach to personalized information retrieval by integrating retrieval augmentation generation (RAG) with a personalized database system. Recent advancements in large language models (LLMs) have shown impressive text generation capabilities but face limitations in knowledge accuracy and hallucinations. Our research addresses these challenges by combining LLMs with structured, personalized data to enhance search precision and relevance. By tagging keywords within personal documents and organizing information into context-based categories, users can conduct efficient searches within their data repositories. We conducted experiments using the GPT-3.5 and text-embedding-ada-002 models and evaluated the RAG assessment framework with five different language models and two embedding models. Our results indicate that the combination of GPT-3.5 and text-embedding-ada-002 is effective for a personalized database question-answering system, with potential for various language models depending on the application. Our approach offers improved accuracy, real-time data updates, and enhanced user experience, making a significant contribution to information retrieval by LLMs and impacting various artificial intelligence applications. Recombination-activating genes (RAGs) play a crucial role in the V(D)J recombination process and the development of immune cells. The development of the immune system and its mechanisms in pigs exhibit greater similarity to those of humans compared to other animals, thus rendering pigs a valuable tool for biomedical research. In this study, we utilized CRISPR/Cas9 gene editing and somatic cell nuclear transfer technology to generate RAG2 knockout (KO) pigs. Furthermore, we evaluated the impact of RAG2 KO on the immune organs and immune cell development through morphological observations, blood analysis and flow cytometry technology. RAG2 KO cell lines were used as donors for cloning. The reconstructed embryos were transplanted into 4 surrogate sows, and after 116 days of gestation, 2 sows gave birth to 12 live piglets, all of which were confirmed to be RAG2 KO. The thymus and spleen sizes of RAG2 KO pigs were significantly smaller than those of wild-type (WT) pigs. Hematoxylin-eosin staining results revealed that the thymus and spleen tissue structures of RAG2 KO pigs were disorganized and lacked the characteristic structures, indicating that RAG2 KO leads to dysplasia of the thymus and spleen. Hematological analysis demonstrated that the total number of white blood cells and lymphocytes in the circulation of RAG2 KO pigs was significantly lower, while the number of eosinophils was higher. Flow cytometry results indicated that the proportions of mature T and B lymphocytes were significantly reduced compared to WT pigs. These findings successfully verified the immunodeficiency phenotype of RAG2 KO pigs. This study may provide experimental animals for the development of tumor models and humanized animals. Mycobacterium tuberculosis (Mtb), a successful human pathogen, resides in host sentinel cells and combats the stressful intracellular environment induced by reactive oxygen and nitrogen species during infection. Mtb employs several evasion mechanisms in the face of the host as a survival strategy, including detoxifying enzymes as short-chain dehydrogenases/reductases (SDRs) to withstand host-generated insults. In this study, using specialized transduction, we have generated a Rv0687 deletion mutant and its complemented strain and investigated the functional role of Rv0687, a member of SDRs family genes in Mtb pathogenesis. A wildtype (WT) and a mutant Mtb strain lacking Rv0687 (RvΔ0687) were tested for the in vitro stress response and in vivo survival in macrophages and mice models of infection. The study demonstrates that the deletion of Rv0687 elevated the sensitivity of Mtb to oxidative and nitrosative stress-inducing agents. Furthermore, the lack of Rv0687 compromised the survival of Mtb in primary bone marrow macrophages and led to an increase in the levels of the secreted proinflammatory cytokines TNF-α and MIP-1α. Interestingly, the growth of WT and RvΔ0687 was similar in the lungs of infected immunocompromised mice; however, a significant reduction in RvΔ0687 growth was observed in the spleen of immunocompromised Rag−/− mice at 4 weeks post-infection. Moreover, Rag−/− mice infected with RvΔ0687 survived longer compared to those infected with the WT Mtb strain. Additionally, we observed a significant reduction in the bacterial burden in the spleens and lungs of immunocompetent C57BL/6 mice infected with RvΔ0687 compared to those infected with complemented and WT Mtb strains. Collectively, this study reveals that Rv0687 plays a role in Mtb pathogenesis. Full article

In order to maintain the polio eradication status, it has become evident that the surveillance of cases with acute flaccid paralysis and of environmental samples must be urgently supplemented with the surveillance of poliovirus excretions among individuals with inborn errors of immunity (IEI). All children with IEI were screened for the excretion of poliovirus during a collaborative study conducted by the ICMR-National Institute of Virology, Mumbai Unit, ICMR-National Institute of Immunohaematology, and World Health Organization, India. A seven-month -old male baby who presented with persistent pneumonia and lymphopenia was found to have severe combined immune deficiency (SCID) due to a missense variant in the RAG1 gene. He had received OPV at birth and at 20 weeks. Four stool samples collected at 4 weekly intervals yielded iVDPV type 1. The child’s father, an asymptomatic 32-year-old male, was also found to be excreting iVDPV. A haploidentical hematopoietic stem cell transplant was performed, but the child succumbed due to severe myocarditis and pneumonia three weeks later. We report a rare case of transmission of iVDPV from an individual with IEI to a healthy household contact, demonstrating the threat of the spread of iVDPV from persons with IEI and the necessity to develop effective antivirals. Recombinase-activating gene (RAG)-deficient SCID patients lack B and T lymphocytes due to the inability to rearrange immunoglobulin and T cell receptor genes. The two RAG genes act as a required dimer to initiate gene recombination. Gene therapy is a valid treatment alternative for RAG-SCID patients who lack a suitable bone marrow donor, but developing such therapy for RAG1/2 has proven challenging. Using a clinically approved lentiviral vector with a codon-optimized RAG1 gene, we report here preclinical studies using CD34+ cells from four RAG1-SCID patients. We used in vitro T cell developmental assays and in vivo assays in xenografted NSG mice. The RAG1-SCID patient CD34+ cells transduced with the RAG1 vector and transplanted into NSG mice led to restored human B and T cell development. Together with favorable safety data on integration sites, these results substantiate an ongoing phase I/II clinical trial for RAG1-SCID. Full article

Two stranded Lepidochelys-like sea turtles were rescued from the Thai Andaman Sea coastline by veterinarians of the Phuket Marine Biological Center (PMBC), one in May of 2019 and another in July of 2021. They were first identified as olive ridley turtles (Lepidochelys olivacea), as the external appearance of both turtles was closer to that species than the other four species found in the Thai Andaman Sea. In fact, when carefully examined, an unusual pattern of the lateral scutes on each turtle was observed, specifically symmetric 5/5 and asymmetric 5/6, both of which are considered rare for L. olivacea and had never been reported in the Thai Andaman Sea. In contrast, this characteristic was more common for the closely related species, Kemp’s ridley (L. kempii), although this species is not distributed in the Indo-Pacific Ocean. Thus, we further investigated their genetic information to confirm species identification using two molecular markers, namely the mtDNA control region and nDNA RAG2. The results from the mtDNA control region sequences using the Basic Local Alignment Search Tool (BLAST) indicated that both individuals exhibited a higher percent identity with L. olivacea (99.81–100.00%) rather than L. kempii (94.29–95.41%) or any other species. A phylogenetic tree confirmed that these two turtles belonged to the L. olivacea clade. Moreover, the results of RAG2 also supported the mtDNA result, as both individuals shared the same RAG2 haplotype with L. olivacea. Thus, we have concluded that the two turtles with unusual lateral scute patterns exhibited genetic consistency with their original species, L. olivacea, which has brought attention to the importance of exploring rare phenotypes in sea turtle populations residing in Thai Seas. There is a pressing need for healthcare conversational agents with domain-specific expertise to ensure the provision of accurate and reliable information tailored to specific medical contexts. Moreover, there is a notable gap in research ensuring the credibility and trustworthiness of the information provided by these healthcare agents, particularly in critical scenarios such as medical emergencies. Pilgrims come from diverse cultural and linguistic backgrounds, often facing difficulties in accessing medical advice and information. Establishing an AI-powered multilingual chatbot can bridge this gap by providing readily available medical guidance and support, contributing to the well-being and safety of pilgrims. In this paper, we present a comprehensive methodology aimed at enhancing the reliability and efficacy of healthcare conversational agents, with a specific focus on addressing the needs of Hajj pilgrims. Our approach leverages domain-specific fine-tuning techniques on a large language model, alongside synthetic data augmentation strategies, to optimize performance in delivering contextually relevant healthcare information by introducing the HajjHealthQA dataset. Additionally, we employ a retrieval-augmented generation (RAG) module as a crucial component to validate uncertain generated responses, which improves model performance by 5%. Moreover, we train a secondary AI agent on a well-known health fact-checking dataset and use it to validate medical information in the generated responses. Our approach significantly elevates the chatbot’s accuracy, demonstrating its adaptability to a wide range of pilgrim queries. We evaluate the chatbot’s performance using quantitative and qualitative metrics, highlighting its proficiency in generating accurate responses and achieve competitive results compared to state-of-the-art models, in addition to mitigating the risk of misinformation and providing users with trustworthy health information. Along with the standard therapies for glioblastoma, patients are commonly prescribed trimethoprim-sulfamethoxazole (TMP-SMX) and dexamethasone for preventing infections and reducing cerebral edema, respectively. Because the gut microbiota impacts the efficacy of cancer therapies, it is important to understand how these medications impact the gut microbiota of patients. Using mice that have been colonized with human microbiota, this study sought to examine how TMP-SMX and dexamethasone affect the gut microbiome. Two lines of humanized microbiota (HuM) Rag1−/− mice, HuM1Rag and HuM2Rag, were treated with either TMP-SMX or dexamethasone via oral gavage once a day for a week. Fecal samples were collected pre-treatment (pre-txt), one week after treatment initiation (1 wk post txt), and three weeks post-treatment (3 wk post txt), and bacterial DNA was analyzed using 16S rRNA-sequencing. The HuM1Rag mice treated with TMP-SMX had significant shifts in alpha diversity, beta diversity, and functional pathways at all time points, whereas in the HuM2Rag mice, it resulted in minimal changes in the microbiome. Likewise, dexamethasone treatment resulted in significant changes in the microbiome of the HuM1Rag mice, whereas the microbiome of the HuM2Rag mice was mostly unaffected. The results of our study show that routine medications used during glioblastoma treatment can perturb gut microbiota, with some microbiome compositions being more sensitive than others, and these treatments could potentially affect the overall efficacy of standard-of-care therapy. A typhoon disaster is a common meteorological disaster that seriously impacts natural ecology, social economy, and even human sustainable development. It is crucial to access the typhoon disaster information, and the corresponding disaster prevention and reduction strategies. However, traditional question and answering (Q&A) methods exhibit shortcomings like low information retrieval efficiency and poor interactivity. This makes it difficult to satisfy users’ demands for obtaining accurate information. Consequently, this work proposes a typhoon disaster knowledge Q&A approach based on LLM (T5). This method integrates two technical paradigms of domain fine-tuning and retrieval-augmented generation (RAG) to optimize user interaction experience and improve the precision of disaster information retrieval. The process specifically includes the following steps. First, this study selects information about typhoon disasters from open-source databases, such as Baidu Encyclopedia and Wikipedia. Utilizing techniques such as slicing and masked language modeling, we generate a training set and 2204 Q&A pairs specifically focused on typhoon disaster knowledge. Second, we continuously pretrain the T5 model using the training set. This process involves encoding typhoon knowledge as parameters in the neural network’s weights and fine-tuning the pretrained model with Q&A pairs to adapt the T5 model for downstream Q&A tasks. Third, when responding to user queries, we retrieve passages from external knowledge bases semantically similar to the queries to enhance the prompts. This action further improves the response quality of the fine-tuned model. Finally, we evaluate the constructed typhoon agent (Typhoon-T5) using different similarity-matching approaches. Furthermore, the method proposed in this work lays the foundation for the cross-integration of large language models with disaster information. Natural Killer Cells Do Not Attenuate a Mouse-Adapted SARS-CoV-2-Induced Disease in Rag2−/− Mice

This study investigates the roles of T, B, and Natural Killer (NK) cells in the pathogenesis of severe COVID-19, utilizing mouse-adapted SARS-CoV-2-MA30 (MA30). To evaluate this MA30 mouse model, we characterized MA30-infected C57BL/6 mice (B6) and compared them with SARS-CoV-2-WA1 (an original SARS-CoV-2 strain) infected K18-human ACE2 (K18-hACE2) mice. We found that the infected B6 mice developed severe peribronchial inflammation and rapid severe pulmonary edema, but less lung interstitial inflammation than the infected K18-hACE2 mice. These pathological findings recapitulate some pathological changes seen in severe COVID-19 patients. Using this MA30-infected mouse model, we further demonstrate that T and/or B cells are essential in mounting an effective immune response against SARS-CoV-2. This was evident as Rag2−/− showed heightened vulnerability to infection and inhibited viral clearance. Conversely, the depletion of NK cells did not significantly alter the disease course in Rag2−/− mice, underscoring the minimal role of NK cells in the acute phase of MA30-induced disease. Together, our results indicate that T and/or B cells, but not NK cells, mitigate MA30-induced disease in mice and the infected mouse model can be used for dissecting the pathogenesis and immunology of severe COVID-19. Cognitive and Motor Capacities Are Poorly Correlated with Agility in Early Pubertal Children: Gender-Stratified Analysis

This research aimed to identify relations of cognitive and power capacities with reactive agility in pubescent boys (n = 55) and girls (n = 46). Cognitive abilities were evaluated by the Stroop test, while the BlazePod system was used to evaluate agility performance conducting 20 yard shuttle and triangle tests of non-reactive (TCODS) and reactive agility (TRAG), respectively. Performance in jumping power was assessed through the squat jump (SJ), countermovement jump (CMJ), and drop jump (DHJ) utilising the Opto Jump system (Microgate, Bolzano, Italy), while sprinting ability over distances of 10 and 20 m was measured using a photocells system. A principal component was extracted from the four Stroop test variables using factor analysis. Forward stepwise multiple regression analysis was conducted separately for boys and girls to evaluate the multivariate relationships among the predictors and the criterion. Among boys, 80% of the TRAG variance was explained (MultipleR = 0.9), with TCODS and SJ as significant predictors (β = 0.53 and −1.01, respectively). For girls, the TCODS was the significant predictor (β = 0.65), explaining 43% of the variance (MultipleR = 0.65). These results show that (i) cognitive abilities measured with the Stroop test were not a reliable tool for predicting TRAG, (ii) jumping power was a significant predictor of TRAG in boys, and (iii) TCODS was a significant predictor of TRAG in girls. The findings indicated that cognitive abilities do not significantly influence reactive agility in pubescent children. It seems that power features have a greater influence on reactive agility, particularly in boys who have more developed motor skills at this age compared to girls. Full article
