Blockchain technology emerged with the advent of Bitcoin and rapidly developed over the past few decades, becoming widely accepted and known by the public. However, in the past decades, the massive adoption of blockchain technology has yet to come. Rather than the scalability issue, the blockchain application is challenged by its expensive usage cost. However, the high cost of blockchain usage is deeply connected with the blockchain consensus and security mechanism. The permissionless blockchain must maintain its high cost for security against the 51% Attack. Chain users indirectly cover the cost as coins are appointed for blockchain usage fees. This conflict prevents the massive adoption of blockchain. Thus, blockchain must be improved to solve those problems: 1. The cost of blockchain usage should be low enough. 2. The blockchain should remain decentralized. 3. The scalability of blockchain must meet the demand.
In my thesis, new approaches are applied to solve the issues above. The key contribution is the discovery of the useful PoW. It extends the Nakamoto PoW with another usage of file data encoding during the same Nakamoto Consensus computation to prove honest data preservation. Based on this theory, a permissionless storage network is proposed as the new security engine for the blockchain. It bridges the high blockchain security cost to the storage users with real demands who are willing to pay for the storage resource. On the other hand, the chain users can benefit from the low transaction fee. Meanwhile, we also provide a scalability solution to shard the blockchain. It enables high TPS and keeps decentralization. The solutions in this thesis provide the answers to all the dependencies of the massive adoption.

Objective. Limited access to breast cancer diagnosis globally leads to delayed treatment. Ultrasound, an effective yet underutilized method, requires specialized training for sonographers, which hinders its widespread use. Approach. Volume sweep imaging (VSI) is an innovative approach that enables untrained operators to capture high-quality ultrasound images. Combined with deep learning, like convolutional neural networks (CNNs), it can potentially transform breast cancer diagnosis, enhancing accuracy, saving time and costs, and improving patient outcomes. The widely used UNet architecture, known for medical image segmentation, has limitations, such as vanishing gradients and a lack of multi-scale feature extraction and selective region attention. In this study, we present a novel segmentation model known as Wavelet_Attention_UNet (WATUNet). In this model, we incorporate wavelet gates (WGs) and attention gates (AGs) between the encoder and decoder instead of a simple connection to overcome the limitations mentioned, thereby improving model performance. Main results. Two datasets are utilized for the analysis. The public "Breast Ultrasound Images" (BUSI) dataset of 780 images and a VSI dataset of 3818 images. Both datasets contained segmented lesions categorized into three types: no mass, benign mass, and malignant mass. Our segmentation results show superior performance compared to other deep networks. The proposed algorithm attained a Dice coefficient of 0.94 and an F1 score of 0.94 on the VSI dataset and scored 0.93 and 0.94 on the public dataset, respectively.

In random sample consensus (RANSAC), the problem of ellipsoid fitting can be formulated as a problem of minimization of point-to-model distance, which is realized by maximizing model score. Hence, the performance of ellipsoid fitting is affected by distance metric. In this paper, we proposed a novel distance metric called the axial distance, which is converted from the algebraic distance by introducing a scaling factor to solve nongeometric problems of the algebraic distance. There is complementarity between the axial distance and Sampson distance because their combination is a stricter metric when calculating the model score of sample consensus and the weight of the weighted least squares (WLS) fitting. Subsequently, a novel sample-consensus-based ellipsoid fitting method is proposed by using the combination between the axial distance and Sampson distance (CAS). We compare the proposed method with several representative fitting methods through experiments on synthetic and real datasets. The results show that the proposed method has a higher robustness against outliers, consistently high accuracy, and a speed close to that of the method based on sample consensus.

Deep learning methods have achieved impressive performance for multi-class medical image segmentation. However, they are limited in their ability to encode topological interactions among different classes (e.g., containment and exclusion). These constraints naturally arise in biomedical images and can be crucial in improving segmentation quality. In this paper, we introduce a novel topological interaction module to encode the topological interactions into a deep neural network. The implementation is completely convolution-based and thus can be very efficient. This empowers us to incorporate the constraints into end-to-end training and enrich the feature representation of neural networks. The efficacy of the proposed method is validated on different types of interactions. We also demonstrate the generalizability of the method on both proprietary and public challenge datasets, in both 2D and 3D settings, as well as across different modalities such as CT and Ultrasound. The development of blockchain applications increased the demand for blockchain performance. Among the attempts of many blockchain scale-out solutions, sharding can improve performance and reduce the storage requirements of each node. Sharding enhances the throughput of the entire blockchain. Most sharding solutions have a fixed number of shards. We propose Wider chain with sharding by account. The number of shards is increased to unlimited. Meanwhile, Wider combines the idea of rollup and retains the main chain to confirm the status of the subchains. This design can avoid the Proof of Work computing power being scattered to the subchains and ensure the security of the blockchain.

Sign language translation (SLT), which generates text in a spoken language from visual content in a sign language, is important to assist the hard-of-hearing community for their communications. Inspired by neural machine translation (NMT), most existing SLT studies adopted a general sequence to sequence learning strategy. However, SLT is significantly different from general NMT tasks since sign languages convey messages through multiple visual-manual aspects. Therefore, in this paper, these unique characteristics of sign languages are formulated as hierarchical spatio-temporal graph representations, including high-level and fine-level graphs of which a vertex characterizes a specified body part and an edge represents their interactions. Particularly, high-level graphs represent the patterns in the regions such as hands and face, and fine-level graphs consider the joints of hands and landmarks of facial regions. To learn these graph patterns, a novel deep learning architecture, namely hierarchical spatio-temporal graph neural network (HST-GNN), is proposed. Graph convolutions and graph self-attentions with neighborhood context are proposed to characterize both the local and the global graph properties. Experimental results on benchmark datasets demonstrated the effectiveness of the proposed method.

The blockchain technology has been changing our daily lives since Bitcoin - i.e., the first decentralized cryptocurrency - was invented and released as open-source software by an unidentified person or a group called Satoshi Nakamoto in 2009. Of many applications which can be implemented based on the blockchain, storage is an important one, a notable example of which is the InterPlanetary File System (IPFS). IPFS is a distributed web based on a peer-to-peer hypermedia protocol to make the web faster, safer, and more open and focuses on public accessible files. To provide a solution for private file storage in the blockchain way, in this paper we propose a Merkle-tree-based File System (MTFS). In MTFS, the blockchain is more than a trust machine; it is an abstract of a cluster system. Distributed random nodes form a tree network cluster without a central controller to provide a secure private storage service and faster message propagation. Advance proxy re-encryption algorithm is applied to guarantee secure file exchanges under permission. Merkle tree will make sure that the files are distributed among the service nodes in a balanced way. The proposed MTFS can be used not only for personal file storage and exchange but also for industry requiring mutual trust in file uploading and downloading in making contracts like insurances.

In recent years, with the rapid development and popularization of BitCoin, the research of blockchain technology has also shown growth. It has gradually become a new generation of distributed, non-centralized and trust-based technology solution. However, the blockchain operation is expensive and transaction is delayed. Take BitCoin as an example. On the one hand, a block is produced every ten minute. On the other hand, once the new block is generated, it takes a certain time to propagate world wide. The slow speed of propagation determines that BitCoin can not use too small block interval time. Ethereum also faces similar problems, so the concept of uncle block was introduced to reduce blockchain forks. This paper introduces a new tree structure based broadcast propagation routing model, providing a novel method to organize network nodes and message propagation mechanism. In oder to avoid the single node failure problem, the tree cluster routing is proposed. The research shows that the tree based routing can accelerate broadcast convergence time and reduce redundant traffic.

Blockchain technology is ushering in another break-out year, the challenge of blockchain still remains to be solved. This paper analyzes the features of Bitcoin and Bitcoin-NG system based on blockchain, proposes an improved method of implementing blockchain systems by replacing the structure of the original chain with the graph data structure. It was named GraphChain. Each block represents a transaction and contains the balance status of the traders. Additionally, as everyone knows all the transactions in Bitcoin system will be baled by only one miner that will result in a lot of wasted effort, so another way to improve resource utilization is to change the original way to compete for miner to election and parallel mining. Researchers simulated blockchain with graph structure and parallel mining through python, and suggested the conceptual new graph model which can improve both capacity and performance.

In this paper, we study the interplay between the epidemic spreading and the diffusion of awareness in multiplex networks. In the model, an infectious disease can spread in one network representing the paths of epidemic spreading (contact network), leading to the diffusion of awareness in the other network (information network), and then the diffusion of awareness will cause individuals to take social distances, which in turn affects the epidemic spreading. As for the diffusion of awareness, we assume that, on the one hand, individuals can be informed by other aware neighbors in information network, on the other hand, the susceptible individuals can be self-awareness induced by the infected neighbors in the contact networks (local information) or mass media (global information). Through Markov chain approach and numerical computations, we find that the density of infected individuals and the epidemic threshold can be affected by the structures of the two networks and the effective transmission rate of the awareness. However, we prove that though the introduction of the self-awareness can lower the density of infection, which cannot increase the epidemic threshold no matter of the local information or global information. Our finding is remarkably different to many previous results--local information based behavioral response can alter the epidemic threshold.
