Team Studer was able to model, in #cell culture, the advanced inclusion pathologies in the human #brain, and show with extensive characterization in postmortem brain that the models capture disease-relevant biology. The inclusionopathy models provide clues to how distinct types of inclusions form, interact with each other, and differentially affect neuronal survival. Combining genome-scale proteomic and genomic screens, the team identified key vesicle trafficking and actin cytoskeleton proteins that are sequestered within a specific inclusion subtype, enabling the group to distinguish among distinct inclusion types in patient postmortem brain. In their recent preprint, Team Edwards investigated how interactions between GABAA receptors (GABAARs) and nicotinic acetylcholine receptors influence dopaminergic axon excitability. They found that GABAARs located on dopaminergic axons regulate the integration of nicotinic input to shape presynaptic excitability in the striatum.
