Exopolysaccharides from the Green Microalga Strain Coelastrella sp. BGV—Isolation, Characterization, and Assessment of Anticancer Potential

Algal metabolites have been extensively studied as potential anticancer therapeutics. Among them, polysaccharides have attracted much attention because of their beneficial biological effects and safety. In the present research, the chemical characteristics, antitumor, and proapoptotic activities of extracellular polysaccharides (EPS) isolated from a new Bulgarian strain of the green microalga Coelastrella sp. BGV were investigated. A fast and convenient method of precipitation with cold ethanol was used to isolate EPS from the culture medium. The chemical characteristics of the isolated EPS were examined by colorimetric and spectrophotometric analyses, HPSEC-RID and HPLC-UV chromatography, and FT-IR spectroscopy. The results showed that the isolated EPS sample consists of three carbohydrate fractions with different molecular weights (11.5 × 104 Da, 30.7 × 104 Da, and 72.4 × 104 Da, respectively) and contains 7.14 (w/w%) protein. HPLC-UV analysis revealed the presence of galactose and fucose. The total uronic acid content in the sample was 4.5 (w/w%). The IR-FT spectrum of EPS revealed the presence of various functional groups typical of a polysaccharide (or proteoglycan) composed primarily of neutral sugars. The anticancer potential of the obtained EPS was assessed using cell lines with cancerous and non-cancerous origins as in vitro experimental models. The results of the performed MTT assay showed that EPS reduced the viability of the cervical and mammary carcinoma cell lines HeLa and MCF-7, while the control non-cancer cell lines BALB/3T3 and HaCaT were less affected. The HeLa cell line showed the highest sensitivity to the effects of EPS and was therefore used for further studies of its anticancer potential. The ability of EPS to inhibit cancer cell migration was demonstrated by wound-healing (scratch) assay. The cell cycle FACS analysis indicated that the EPS treatment induced significant increases in the sub G1 cell population and decreases of the percentages of cells in the G1, S, and G2-M phases, compared to the control. The fluorescent microscopy studies performed using three different staining methods in combination with Annexin V-FITC flow cytometric analysis clearly demonstrate the ability of EPS to induce cancer cell death via the apoptosis pathway. Moreover, an altered pattern and intensity of the immunocytochemical staining for the apoptosis- and proliferation-related proteins p53, bcl2, and Ki67 was detected in EPS-treated HeLa cancer cells as compared to the untreated controls. The obtained results characterize the new local strain of green microalgae Coelastrella sp. BGV as a producer of EPS with selective antitumor activity and provide an opportunity for further studies of its pharmacological and biotechnological potential. Full article

Changes of Antioxidant and Functional Components in Various Salt-Aged and Fresh Radishes after Fermentation

Many studies have found that salted radishes offer various health benefits, such as enhancing antioxidant levels and increasing GABA. This study fermented a mixture of 20-year-old salted radishes (20-S. radishes), 2-year-old salted radishes (2-S. radishes), 20-year-old salted radishes combined with fresh radishes (R + 20-radishes), and fresh radishes with eight whole grains fermentation as a starter (EGS) for 8 weeks. EGS was derived from the saccharified fermentation of millet, wheat, sorghum, black rice, buckwheat, pearled rice, black glutinous rice, and quinoa, serving as a carbon source for microorganisms and replacing the traditional sugar-based fermentation method. During the fermentation process, the bacterial count of the 20-year-old salted radishes significantly increased to 11.08 ± 0.03 log CFU/mL, which was much higher than the other three groups. Pichia manshurica LYC1722 was identified in all four groups after isolation. After 8 weeks of fermentation, 20-S. radishes showed the highest concentrations of gamma-aminobutyric acid (GABA) and glucuronic acid in functional components, at 18.40 ± 0.69 ppm and 14,162.84 ± 48.22 ppm, respectively. In terms of antioxidant components, 20-S. radishes exhibited a total phenolic content (TPC) and total flavonoid content (TFC) of 0.81 ± 0.01 mg/mL and 42.78 ± 0.60 mg/L, respectively. Regarding antioxidant capability, 20-S. radishes displayed ABTS radical scavenging activity and DPPH radical scavenging activity at 184.42 ± 0.28 μg/mL and 9.13 ± 0.28 μg/mL, respectively. These values were the highest among the four groups evaluated. Fresh radishes exhibited the highest angiotensin-converting enzyme (ACE) inhibition after fermentation among the four groups, reaching 69.04 ± 2.82%, slightly higher than 20-S. radishes. These results show that 20-S. radishes are expected to become a novel health beverage in the future. Full article

In this study, in order to verify the effects due to the addition of spirulina (Arthrospira platensis) in a food product, a wildflower honey was analyzed in terms of chemical composition, physicochemical properties and antioxidant activity before and after the addition of the spirulina. HS-SPME/GC–MS and HPLC/UV were applied to carry out the chemical analyses. The obtained results demonstrated that the volatile profile and also the sugar content were significantly influenced by the addition of spirulina, showing significant qualitative and quantitative differences compared to honey without spirulina. The increase in HMF in honey added with spirulina was significant, demonstrating that its presence could accelerate the Maillard reaction. Electrical conductivity measured by using a conductometer was also increased while the moisture content was reduced in honey enriched with spirulina. Instead, the pH value was similar between the two samples. On the other hand, honey fortification with spirulina determined a significant increase of 12.5% in the total phenolic content (TPC), and a 56.25% increase in protein content. Further, the total antioxidant capacity (TAC) was also evaluated and a significant increase was determined as a result of the addition of spirulina. In conclusion, honey enriched with A. platensis was found to be characterized by a high pool of bioactive metabolites as well as significant changes in almost all the measurements performed. Background: Despite clinical interventional studies on the influence of diet on periodontal inflammatory parameters, there has been no practice-based cross-sectional study from a German population to date that has conducted both a comprehensive dental and periodontal examination and a thorough validated assessment of dietary behavior. Therefore, the aim of this pilot study was to evaluate, in a proof of concept, whether there is a correlation between the overall periodontal inflammatory surface area (PISA), periodontal clinical parameters (pocket probing depths (PPD), clinical attachment loss (CAL), bleeding on probing (BOP), furcation involvement (FI), tooth mobility (TM)), and the dietary behavior of patients with periodontal disease when utilizing a practice-based research network. The primary outcome was the correlation between the periodontal inflammatory surface (PISA) and the dietary assessment data. Materials and Methods: The practice-based research network, consisting of eight Master’s graduates, recruited patients who met the inclusion and exclusion criteria and performed a periodontal examination together with the assessment of dietary behavior using a digital version of the validated retrospective dietary recall (DEGS/RKI). Statistical analyses included linear regression models adjusted for age and smoking and unpaired t-tests, conducted using STATA 17.0 with a significance level of 5%. In addition, the data obtained were classified according to the currently recommended amounts of daily intake. Results: A total of 1283 teeth were analyzed, with 60.25% (773 teeth) requiring treatment. The average PISA was 753.16 mm2 (SD ± 535.75 mm). Based on dietary guidelines, the studied population consumed excessive amounts of extrinsic sugars and fats, while their fiber and legume intake was insufficient. The intake of certain nutrients, including water-soluble fibers, specific fatty acids, vitamins (D, B1, B2, B6, and B12), iron, and zinc, was associated with reduced PISA, PPD, CAL, and BOP. Conclusion: Within the limits of the current study, including its cross-sectional design and cohort size, the outcomes demonstrated the influence of nutrition on periodontal health. Full article

Longan (Dimocarpus longan Lour.) is an important tropical and subtropical fruit, and most of its cultivars bloom once a year (once-flowering, OF). Dimocarpus longan cv. ‘Sijimi’ (SJ) is a tropical ecotype variety that blooms several times a year (continuous-flowering, CF) without the need for low-temperature induction. Several studies have focused on the mechanism of continuous flowering in SJ longan; however, none used leaves as research material. As leaves are a key organ in sensing floral-induction signals, we compared gene-expression differences between mature leaves of CF (SJ) and OF (D. longan cv. ‘Shixia’ (SX) and D. longan cv. ‘Chuliang’ (CL)) longan by transcriptome sequencing. An average of 47,982,637, 43,833,340 and 54,441,291 clean reads were obtained for SJ, SX and CL respectively, and a total of 6745 differentially expressed genes (DEGs) were detected. Following Metabolic pathways, Plant-pathogen interaction and Biosynthesis of secondary metabolites, most of the other genes were assigned to the KEGG classifications of MAPK signaling pathway- plant, Plant hormone signal transduction, Amino sugar and nucleotide sugar metabolism and Starch and sucrose metabolism. WGCNA analysis clustered genes into 27 modules, among which bisque4 and darkorange2 module genes specifically were expressed at low and high levels in SJ, respectively. Different gene-expression patterns were detected between CF and OF longan in bisque4 and darkorange2 modules, especially the high levels of transcription factor (TF) expression and the large number of gibberellic acid (GA)-signaling-pathway-specific genes expressed at high levels in CF longan (SJ). Floral-induction-gene expression levels in CF longan, such as levels of GA-signaling-related and FT genes, were always high. In CF longan, after vegetative-growth accumulation, flowers could be directly induced, thereby eliminating the need for low-temperature induction. Full article

Isolation of new plant-derived lactic acid bacteria (LAB) is highly prioritized in developing novel starter cultures for plant-based fermentation. This study explores the diversity of LAB in Danish flowers and their potential use for plant-based food fermentation. A total of 46 flower samples under 34 genera were collected for LAB isolation. By introducing an enrichment step, a total of 61 LAB strains were isolated and identified using MALDI-TOF and 16S rRNA sequencing. These strains represent 24 species across 9 genera, predominantly Leuconostoc mesenteroides, Fructobacillus fructosus, Apilactobacillus ozensis, and Apilactobacillus kunkeei. Phenotypic screening for exo-cellular polysaccharide production revealed that 40 strains exhibited sliminess or ropiness on sucrose-containing agar plates. HPLC analysis confirmed that all isolates produced exo-cellular polysaccharides containing glucose, fructose, or galactose as sugar monomers. Therefore, the strains were glucan, fructan, and galactan producers. The suitability of these strains for plant-based fermentation was characterized by using almond, oat, and soy milk. The results showed successful acidification in all three types of plant-based matrices but only observed texture development in soy by Leuconostoc, Weissella, Lactococcus, Apilactobacillus, and Fructobacillus. The findings highlight the potential of flower-derived LAB strains for texture development in soy-based dairy alternatives. Full article

Updated and accurate crop yield maps play a key role in the agricultural environment. Their application enables the support for sustainable agricultural practices and the formulation of effective strategies to mitigate the impacts of climate change. Farmers can apply the maps to gain an overview of the yield variability, improving farm management practices and optimizing inputs to increase productivity and sustainability such as fertilizers. Earth observation (EO) data make it possible to map crop yield estimations over large areas, although this will remain challenging for specific crops such as sugarcane. Yield data collection is an expensive and time-consuming practice that often limits the number of samples collected. In this study, the sugarcane yield estimation based on a small number of training datasets within smallholder crop systems in the Tha Khan Tho District, Thailand for the year 2022 was assessed. Specifically, multi-temporal satellite datasets from multiple sensors, including Sentinel-2 and Landsat 8/9, were involved. Moreover, in order to generate the sugarcane yield estimation maps, only 75 sampling plots were selected and surveyed to provide training and validation data for several powerful machine-learning algorithms, including multiple linear regression (MLR), stepwise multiple regression (SMR), partial least squares regression (PLS), random forest regression (RFR), and support vector regression (SVR). Among these algorithms, the RFR model demonstrated outstanding performance, yielding an excellent result compared to existing techniques, achieving an R-squared (R2) value of 0.79 and a root mean square error (RMSE) of 3.93 t/ha (per 10 m × 10 m pixel). Furthermore, the mapped yields across the region closely aligned with the official statistical data from the Office of the Cane and Sugar Board (with a range value of 36,000 ton). Finally, the sugarcane yield estimation model was applied to over 2100 sugarcane fields in order to provide an overview of the current state of the yield and total production in the area. In this work, the different yield rates at the field level were highlighted, providing a powerful workflow for mapping sugarcane yields across large regions, supporting sugarcane crop management and facilitating decision-making processes. Introduction: This study aimed to understand the influence of diet and nutrition items on gastrointestinal symptoms in patients with Hirschsprung Disease (HD). Method: An online questionnaire was created to obtain patient-reported outcomes using the multinational Holistic Care in Hirschsprung Disease Network. This was distributed in Dutch, English, German, Italian, Polish, and Swedish via patient associations. Information on demographics, the extension of disease, current diet, and the influence of food ingredients on bowel function were obtained. Results: In total, 563 questionnaires were answered by parents or patients themselves. The length of the aganglionic segment was short in 33%, long in 45%, total colonic aganglionosis (TCA) in 11%, and involved the small intestine in 10%. Overall, 90% reported following a mixed diet, and 31% reported taking probiotics, with twice as many patients taking probiotics in the TCA group compared to standard HD. Mealtimes and behaviours around eating were affected by 61%, while 77% had established food items that worsened symptoms, and of these, 80% stated that they had worked these items out themselves. A high-fibre diet was followed by 24% and 18% a low-fibre diet. Symptoms were reported, particularly from dairy in 30%, fruits in 39%, pulses in 54%, and sugar in 48%. Conclusions: This first multinational survey on diet and bowel function in HD reports an association between certain dietary items with gastrointestinal symptoms. This study can support an improved understanding of the interaction between food items and bowel function in children with HD. We suggest a multidisciplinary approach to balance dietary exclusions and support adequate growth, preventing nutrition deficiencies and enhancing quality of life. Full article

Rosa roxburghii (R. roxburghii), native to the southwest provinces of China, is a fruit crop of important economic value in Guizhou Province. However, the changes in fruit quality and flavor during R. roxburghii fruit ripening have remained unknown. Here, this study investigated the changes of seven active components and volatile organic compounds (VOCs) during the ripening of the R. roxburghii fruit at five different ripening stages including 45, 65, 75, 90, and 105 days after anthesis. The results indicated that during the ripening process, the levels of total acid, vitamin C, and soluble sugar significantly increased (p < 0.05), while the levels of total flavonoids, superoxide dismutase (SOD), and soluble tannin significantly decreased (p < 0.05). Additionally, the content of total phenol exhibited a trend of first decreasing significantly and then increasing significantly (p < 0.05). A total of 145 VOCs were detected by HS-SPME-GC-MS at five mature stages, primarily consisting of aldehydes, alcohols, esters, and alkenes. As R. roxburghii matured, both the diversity and total quantity of VOCs in the fruit increased, with a notable rise in the contents of acids, ketones, and alkenes. By calculating the ROAV values of these VOCs, 53 key substances were identified, which included aromas such as fruit, citrus, green, caramel, grass, flower, sweet, soap, wood, and fat notes. The aromas of citrus, caramel, sweet, and wood were predominantly concentrated in the later stages of R. roxburghii fruit ripening. Cluster heatmap analysis revealed distinct distribution patterns of VOCs across five different maturity stages, serving as characteristic chemical fingerprints for each stage. Notably, stages IV and V were primarily characterized by a dominance of alkenes. OPLS-DA analysis categorized the ripening process of R. roxburghii fruit into three segments: the first segment encompassed the initial three stages (I, II, and III), the second segment corresponded to the fourth stage (IV), and the third segment pertained to the fifth stage (V). Following the variable importance in projection (VIP) > 1 criterion, a total of 30 key differential VOCs were identified across the five stages, predominantly comprising ester compounds, which significantly influenced the aroma profiles of R. roxburghii fruit. By integrating the VIP > 1 and ROAV > 1 criteria, 21 differential VOCs were further identified as key contributors to the aroma changes in R. roxburghii fruit during the ripening process. This study provided data on the changes in quality and aroma of R. roxburghii fruit during ripening and laid the foundation for the investigation of the mechanism of compound accumulation during ripening. WA KP exhibited higher total sugar (16.3 ± 1.0 g/100 g DW) and free ellagic acid (EA) (23.2 ± 1.7 mg/g DW), along with abundant vitamin C (25.20 ± 0.16 to 131.50 ± 0.20 mg/g DW) compared to Northern Territory KP fruits. The fruit showed strong antioxidant activities, α-glucosidase inhibition, and effectiveness against bacteria, with positive correlations to total phenolic content (TPC), vitamin C, and EA. These findings highlight WA KP’s potential for functional foods and pharmaceuticals, emphasizing the importance of TPC, vitamin C, and EA in selecting high-quality fruit. Full article

Strawberry wine production is a considerable approach to solve the problem of the Chinese concentrated harvesting period and the short shelf life of strawberries, but the appropriative strawberry cultivars for fermentation are still undecided. In this study, the strawberry juice and wines of four typical strawberry cultivars named Akihime (ZJ), Sweet Charlie (TCL), Snow White (BX), and Tongzhougongzhu (TZ) were thoroughly characterized for their physicochemical indicators, bioactive compounds, and volatile organic components (VOCs) to determine the optimal strawberry cultivars for winemaking. The results showed that there were significant differences in the total sugar content, pH, total acid, and other physicochemical indexes in the strawberry juice of different cultivars, which further affected the physicochemical indexes of fermented strawberry wine. Moreover, the content of polyphenols, total flavonoids, vitamin C, and color varied among the four strawberry cultivars. A total of 42 VOCs were detected in the strawberry juice and wines using headspace solid-phase microextraction coupled with gas chromatography–mass spectrometry (HS-SPME-GC-MS), and 3-methyl-1-butanol, linalool, trans-2-pinanol, hexanoic acid, and hexanoic acid ethyl ester were the differential VOCs to identify the strawberry wine samples of different cultivars. Overall, strawberry cultivar ZJ had a relatively high VOC and bioactive compound content, indicating that it is the most suitable cultivar for strawberry wine fermentation. In addition to determining the relatively superior fermentation characteristics of cultivar ZJ, the results may provide a theoretical basis for the raw material quality control and quality improvement of strawberry wine. Salinity is a major problem, impeding soil productivity, agricultural sustainability, and food security, particularly in dry regions. This study integrates quinoa, a facultative halophyte, into a pomegranate-based agroforestry with saline irrigation in northeast Morocco. We aim to explore this agroforestry model’s potential in mitigating salinity’s effects on quinoa’s agronomic and biochemical traits and evaluate the land equivalent ratio (LER). Field experiments in 2020 and 2021 used a randomized block design with three replicates, including monocropping and agroforestry systems, two salinity levels (1.12 and 10.5 dS m−1), four quinoa genotypes (Titicaca, Puno, ICBA-Q4, ICBA-Q5), and a pomegranate control. Salinity significantly decreased total dry matter (40.5%), root dry matter (50.7%), leaf dry matter (39.2%), and root-to-shoot ratio (7.7%). The impact was more severe in monoculture than in agroforestry, reducing dry matter (47.6% vs. 30.7%), grain yield (46.3% vs. 26.1%), water productivity (47.5% vs. 23.9%), and total sugar (19.2% vs. 5.6%). LER averaged 1.86 to 2.21, indicating 86–121% higher productivity in agroforestry. LER averaged 1.85 at 1.12 dS m−1 and 2.18 at 10.5 dS m−1, reaching 2.21 with pomegranate-ICBA-Q5 combination. Quinoa–pomegranate agroforestry emerges as an innovative strategy, leveraging quinoa’s salt resistance and agroforestry’s potential to mitigate salinity impacts while enhancing land use efficiency. Bananas are one of the most consumed fruits in the world and are essential for food security and in the agricultural economy. Their sustainable production is vital for many communities. Herein, the effects of banana cultivars, cropping systems (monocropping and intercropping with lemongrass), and the interaction between these factors on the physicochemical characteristics of ripe and unripe fruit were evaluated. The intercropping of bananas with lemongrass did not alter the main quality indicators of the fruit. The ripe fruits of the SCS451 Catarina cultivar stood out for their higher fruit mass, pulp/peel ratio, and firmness, while the fruits of the Prata Anã and BRS FHIA Maravilha cultivars had the highest SS, SS/TA ratio, pH, and reducing and total sugars. The cultivar SCS451 Catarina had the highest mineral content in the unripe fruit, and for all cultivars, the order of mineral levels was K > N > Mg > P > Ca > Fe > Zn. The higher content of total and resistant starch in the unripe fruit of the Prata Anã and BRS FHIA Maravilha cultivars, combined with the characteristics of the ripe fruit, make them suitable for both fresh consumption and industrial use. Diversifying cultivars improves the quality of banana fruit, and intercropping with lemongrass can be a sustainable method of managing banana fields. Full article

This study aimed to use waste figs as an alternative substrate for bacterial cellulose (BC) production by Komagataeibacter xylinus and optimize the identified process parameters to maximize the concentration of BC. Among the nutrients screened by Plackett–Burman (PB) design, yeast extract was found to be significant in BC production. Response surface methodology was used to investigate the effect of fermentation parameters on BC production. A maximum BC concentration of (8.45 g/L), which is among the highest BC concentrations reported so far, was achieved at the optimum levels of fermentation variables (initial pH 6.05, initial sugar concentration 62.75 g/L, temperature 30 °C). The utilization of response surface methodology (RSM) proved valuable in both optimizing and finding the interactions between process variables during BC production. Scanning electron microscope (SEM) analysis showed a dense structure of BC, characterized by ribbon-like nanofibrils with diameters ranging from 23 to 90 nm while the attenuated total reflection–Fourier transform infrared (ATR-FTIR) spectrum of BC confirmed that the material obtained was cellulose. The X-ray diffraction (XRD) analysis showed that the crystallinity of the BC samples was 70% for BC produced on waste fig medium and 61% for BC produced on Hestrin–Schramm (HS) medium. This is the first detailed study on the production of BC from waste figs, and the findings of this study demonstrated that waste figs can be used as an effective substrate for the production of BC. A diet with low content of fermentable oligo-, di-, and monosaccharides and polyols (FODMAP) is established treatment for irritable bowel syndrome (IBS), with well-documented efficiency. A starch- and sucrose-reduced diet (SSRD) has shown similar promising effects. The primary aim of this randomized, non-inferiority study was to test SSRD against low FODMAP and compare the responder rates (RR = ∆Total IBS-SSS ≥ −50) to a 4-week dietary intervention of either diet. Secondary aims were to estimate responders of ≥100 score and 50% reduction; effects on extraintestinal symptoms; saturation; sugar craving; anthropometric parameters; and blood pressure. 155 IBS patients were randomized to SSRD (n = 77) or low FODMAP (n = 78) for 4 weeks, with a follow-up 5 months later without food restrictions. The questionnaires Rome IV, IBS-severity scoring system (IBS-SSS), and visual analog scale for IBS (VAS-IBS) were completed at baseline and after 2 and 4 weeks and 6 months. Weight, height, waist circumference, and blood pressures were measured. Comparisons were made within the groups and between changes in the two groups. There were no differences between groups at baseline. The responder rate of SSRD was non-inferior compared with low FODMAPs at week 2 (79.2% vs. 73.1%; p = 0.661;95% confidence interval (CI) = −20–7.2) and week 4 (79.2% vs. 78.2%; p = 1.000;95%CI = −14–12). Responder rate was still high when defined stricter. All gastrointestinal and extraintestinal symptoms were equally improved (p < 0.001 in most variables). SSRD rendered greater reductions in weight (p = 0.006), body mass index (BMI) (p = 0.005), and sugar craving (p = 0.05), whereas waist circumference and blood pressure were equally decreased. Weight and BMI were regained at follow-up. In the SSRD group, responders at 6 months still had lowered weight (−0.7 (−2.5–0.1) vs. 0.2 (−0.7–2.2) kg; p = 0.005) and BMI (−0.25 (−0.85–0.03) vs. 0.07 (−0.35–0.77) kg/m2; p = 0.009) compared with baseline in contrast to non-responders. Those who had tested both diets preferred SSRD (p = 0.032). In conclusion, a 4-week SSRD intervention was non-inferior to low FODMAP regarding responder rates of gastrointestinal IBS symptoms. Furthermore, strong reductions of extraintestinal symptoms were found in both groups, whereas reductions in weight, BMI, and sugar craving were most pronounced following SSRD. Full article
